Plasmid extraction method
Solution I (for plasmid extraction)
Component concentrations: 25 mM Tris-HCl (pH 8.0), 10 mM EDTA, 50 mM Glucose
Preparation volume: 1 L
Preparation method: 1. Measure the following solutions into a 1 L beaker.
1M Tris-HCl (PH8.0) | 25ml |
0.5M EDTA (PH8.0) | 20ml |
20% Glucose (1.11M) | 45ml |
dH 2 O | 910m |
2. After autoclaving, store at 4°C.
3. Add 2 ml of RNase A (20 mg/ml) to every 50 ml of Solution I before use.
Solution II (for plasmid extraction)
Component concentration: 200mM NaOH, 1% (W/V) SDS
Preparation amount: 500ml Preparation method:
1. Measure the following solutions and place them in a 500ml beaker
10% SDS 50ml
2N NaOH 50ml
2. Add sterilized water to 500ml and mix well
3. Store at room temperature, this solution should not be stored for more than one month
Note: SDS is prone to bubbles, do not stir vigorously
Solution III (for plasmid extraction)
Component concentrations: 3 M KOAc, 5 M CH3COOH
Preparation volume: 500 ml Preparation method:
1. Weigh the following reagents into a 500 ml beaker.
KOAc 147g
CH3COOH 57.5ml
2. Add 300 ml of deionized water and stir to dissolve.
3. Add deionized water to bring the solution to 500 ml.
4. After autoclaving, store at 4°C.
