Recipe 1: beef extract agar medium
0.3g beef extract, 1.0g peptone, 0.5g sodium chloride, 1.5g agar,100ml water
Add 100 ml of water to the beaker, add beef extract, peptone and sodium chloride, mark the outside of the beaker with crayons, and heat it on the fire. After the components in the beaker are dissolved, add the agar, stirring constantly to avoid sticking to the bottom. After the agar is completely dissolved, make up for the loss of water, adjust the pH to 7.2-7.6 with 10% hydrochloric acid or 10% sodium hydroxide, divide them into each test tube, add cotton plugs, and autoclave for 30 minutes.

Recipe 2: Potato medium
Take 250 grams of fresh beef heart (with the fat and blood vessels removed), finely chop it into minced meat with a knife, add 500 ml of distilled water and 5 grams of peptone. Make a mark on the beaker, bring to a boil, and simmer for 2 hours. After filtering, the filtered minced meat is dried, and the pH value of the filtrate is adjusted to about 7.5. Add 10 ml of broth and a small amount of dried beef hearts in a small amount of crushed beef to each test tube, sterilize and set aside.

Recipe 3: Rhizobium culture medium
Glucose 10 g Dipotassium hydrogen phosphate 0.5 g
Calcium carbonate 3 g Magnesium sulfate 0.2 g
Yeast powder 0.4g agar 20g
1000 ml water 1% crystal violet solution 1 ml
First boil the agar with water to dissolve, then add the other components separately, stir to dissolve, pack, sterilize, and set aside.
