Buffer solution configuration for 0.5 M EDTA and 1 M DTT

(1) 0.5M EDTA (pH8.0)
Component concentration: 0.5 M EDTA
Preparation volume: 1 L
Preparation method:
Weigh out 186.1 g of Na2EDTA·2H2O and place it in a 1 L beaker.
2. Add about 800 ml of deionized water and stir well.
3. Adjust pH to 8.0 with NaOH (approx. 20 g NaOH).
Note: EDTA can only dissolve completely when the pH value reaches 8.0.
4. Add deionized water to bring the solution to 1 L.
5. Divide an appropriate amount into small portions and sterilize at high temperature and high pressure.
6. Store at room temperature.

(1) 1 M DTT
Component concentration: 1 M DTT
Preparation volume: 20 ml
Preparation method:
1. Weigh 3.09 g of DTT into a 50 ml plastic centrifuge tube.
2. Add 20 ml of 0.01 M NaOAc (pH 5.2), dissolve and filter through a 0.22 mm filter.
3. Divide an appropriate amount into small portions and store at -20°C.
