3 M sodium acetate and PBS Buffer buffer configuration scheme

1) 3 M sodium acetate (pH 5.2)
Component concentration: 3M sodium acetate
Preparation amount: 100ml
Preparation method:
1. Weigh 40.8g NaAc·3H 2 O into a 100-200ml beaker, add 40ml of deionized water and stir to dissolve
2. Add glacial acetic acid to adjust pH to 5.2
3. Add deionized water to dilute the solution to 100ml
4 Store at room temperature after autoclaving.

2) PBS Buffer
Component concentrations: 137 mM NaCl, 2.7 mM KCl, 10 mM Na2HPO4, 2 mM KH2PO4
Preparation volume: 1 L
Preparation method:
1. Weigh the following reagents into a 1 L beaker.

NaCl 8g
KCl 0.2g
Na 2 HPO 4 1.42g
KH 2 PO 4 0.27g

2. Add about 800 ml of deionized water to the beaker and stir well to dissolve.
3. Add concentrated hydrochloric acid dropwise to adjust the pH to 7.4, then add deionized water to bring the solution to 1 L.
4. Store at room temperature after autoclaving.
Note: There are no divalent cations in the above PBS Buffer. If necessary, 1 mM CaCl2 and 0.5 mM MgCl2 can be supplemented in the formula.
