Formation of amylase during grain seed germination
[principle]
When the seed germinates, the activity of the hydrolase is greatly enhanced, and the organic matter stored in the cotyledon or endosperm is degraded into simple compounds under their action for the needs of seedling growth. Amylase is formed during germination and can hydrolyze starch into sugar. The blue reaction of starch to I-KI can be used to detect the presence of amylase.

[Steps]
1. Take some wheat seeds for germination and set aside.
2. Weigh 2g of agar and place it in a beaker, add 100ml of distilled water, heat on a small fire, and stir constantly to dissolve the agar. Take another 1g of starch in a small beaker, add a little water and mix thoroughly. 3. Take 20 germinated and ungerminated wheat seeds, respectively add 5ml of distilled water to grind them in a mortar, then wash all the grounds in a small beaker with 5ml of distilled water, and let stand for 15 minutes. 4. Dip a small amount of liquid (germinated and ungerminated seed extracts) with a pen, draw a word on the starch agar plate in the glass dish, cover the dish, and place it in the incubator at 25°C.
